Manna Dey was born in Calcutta on May 1, 1919 at Mahamaya and Purna Chandra Dey in a Bengali family. Apart from his parents, his youngest father, Sangeetacharya Krishna Chandra Dey, inspired and impressed him a lot. He received his elementary education in a small primary school, Indu Babar Pathshala. He started doing stage shows at school from 1929. He attended scottish church collegiate school and scottish church college. He participated in sports like wrestling and boxing during his college days, trained from gobar guha. He graduated from Vidyasagar College.

Manna Dey started learning music from Krishna Chandra Dey and Ustad Dabir Khan. During this learning period, he has been ranked first in three different categories of inter-collegiate singing competitions for three consecutive years. In 1942, Dey accompanied Krishna Chandra Dey on a visit to Bombay. There he first started working as an assistant music director under Krishna Chandra Dey, and then under Sachin Dev Burman. Later, he assisted other music creators and then started working independently. Working independently as a music director for various Hindi films, Manna Dey continued to learn music in Hindustani classical music from Ustad Aman Ali Khan and Ustad Abdul Rahman Khan.

Dey started his career in playback singing with the film Tamannaah in 1942. Musical scores Krishna Chandra Dey and Manna sang a duet with Suraiya titled "Jago Aye Usha Ponchi Bole Jaago" which was a quick hit. But it was only in 1943 that he got his first solo break with Ram Rajya. Incidentally, the film's producer Vijay Bhatt and its composer Shankar Rao Vyas had approached K C Day with an offer of playback singing in the film. On June 8, 2013, DEE was admitted to icu at a bengaluru hospital, as chest infection caused other complications. His health gradually improved and about a month later doctors resorted to ventilators. Later, he was released from the hospital. After which he died.
